The Division of Applied Veterinary Research (DAVR) in the FDA Center for Veterinary Medicine, Office of Research, requires scientific equipment to transfer RNA/DNA directly into the nucleus of immortalized cell lines and primary cells/stem cells. Primary and stem cells are notoriously difficult, if not impossible, to transfect using cationic lipid mediated methods (Fugene, Lipofectamine, etc.).
FDA will perform an experimental technique that requires the transfer of RNA and DNA directly into the nucleus of a cell to cause double stranded DNA breaks. This will allow for the introduction of nucleotide insertions or deletions, and for the introduction of exogenous DNA sequences into targeted genomic regions. We will also transfer double stranded oligodeoxynucleotides (dsODNs) into the double stranded breaks in the genome. These dsDNA sequences must be delivered directly into the nucleus of the cell, and cationic lipid mediated methods will not work.
