Optical Illumination System
The USDA, ARS, LSO intends to solicit and negotiate with only one source Cytoviva, 300 North Dean Rd., Suite 5 - PMB 157, Auburn, AL 36830 to purchase an optical illumination system that contains the following: Dual mode (UV and white light) optical illuminator compatible for use with an Olympus BX41 inverted microscope (existing inventory). The system is needed to perform high speed videography simultaneously using UV and white light. The system must include:
• Compatibility with the observation of live protozoa and bacteria.
• Darkfield microscope illumination system with integrated dual mode transmitted light fluorescence.
• Video camera capable of up to 100 frames per second.
• Darkfield illumination system containing a 1.2-1.4 NA (numerical aperture) cardioid oil condenser, capable of accepting light directly via 3mm ID, liquid light guide and producing automated internalized pre-aligned Koehler illumination and the main feature of critical illumination.
• Excitation filters: DAPI (4',6-diamidino-2-phenylindole blue stain), FITC (fluorescein isothiocyanate green stain), TX Red (sulforhodamine 101 acid chloride red stain) and a triple pass excitation filter, plus a triple pass DAPI, FITC, TX Red emission filter.
• A liquid light connection must be available to remove heat source from sample.
• The system must include a UPL (Universal Plan) Fluorite (objective) 60X .65-1.25 NA with iris that will be compatible with an Olympus BX-41 microscope.
• A Xenon illuminated Fiber Optic Strobe capable of synchronization with the high speed camera.
• A high frame rate color digital camera with the following features:
o Scientific CMOS (sCMOS) sensor
o Resolution (h x v) 2560 x 2160 pixel or better
o Shutter modes rolling shutter (RS) with free selectable readouts
o Global/snapshot shutter (GS)
o Dynamic range 27,000:1 (88.6 dB)
o Quantum efficiency > 60%
o SMA connectors
o Data interface Camera Link Full (10 taps, 85 Mhz)
o Time stamp in image (1 us resolution)
o 100 fps @ RS, fast scan or better
o 50 fps @ GS, fast scan or better
o Exposure/ shutter time range between 500 us..2 s RS or better
o Dynamic range A/D 16 bit
