The single-cell genomics is one of the cutting-edge technologies developed in 2015, and it has generated lots of research interests within NIH, as well as the research community worldwide. We have utilized this new technology to improve and facilitate our current and future projects and clinical trials. Specifically, we have used single-cell RNAseq data to identify T cell receptor sequences, which will be utilized for highly-personalized T cell therapy targeting mutated antigens in cancer patients. We have also used single-cell RNAseq data to understand T cell biology at the single-cell level in our clinical trials. The results will help us further improve our T-cell based immunotherapies in cancer patients. Lastly, we have utilized single-cell whole-exome sequencing data to identify mutations in circulating tumor cells, and to evaluate tumor heterogeneity in our tumor samples from our clinical trials. Therefore, these two instruments are essential for our current and ongoing research projects.
